Heavy metal Plumbum (Pb) poisoning that occurs in children recently is the
main problem in developed countries as well as in developing countries. Many
researches about heavy metal Plumbum (Pb) have shown that children are more susceptible to negative
influences of heavy metal spread than adults. Polluted environment is certainly
influential to the spread of plumbum at children which can be seen from DNA
genome coding Amino Acid Levulinat Dehidratase (ALAD) enzyme. This research is
aimed at getting optimum ALAD gene fragment which can be used as environmental
Pb polluted gene mark. The method used in the research is descriptive method
with laboratory PCR AAS and RFLP as well as conducted interviews to obtain
information of research subject. 30 research subjects consisting 10 students of
SD Pisangan Baru 01, 10 students of SD Pulo Gebang 01, and 10 students of SD
Malaka Jaya 10 are used as research samples. The results show that 4 from 30
research samples succeed to be amplified by ALAD gene fragment along 500bp
which represent each elementary school. The AAS test of school hedgerow
resulting 2 bends in RFLP test along 150 bp and 350 bp proves that there is
plumbum (Pb) contamination in environment at Amino Acid Levulinat Dehydratase
(ALAD) Gene Mutation.
